摘要:慢性乙型肝炎,其病原为Hepatitis B virus,简称HBV.中国作为HBV感染人数最多的国家,其中有二千多万患者被确诊为慢性肝炎(CHB),而CHB是导致肝硬化和肝癌的最主要诱因.中国科学院武汉病毒研究所研究员胡康洪带领着他的学科组,致力于慢性乙型肝炎病毒的复制机理及抗病毒治疗领域的深入研究 ……
慢性乙型肝炎,其病原为Hepatitis B virus,简称HBV。中国作为HBV感染人数最多的国家,其中二千多万确诊为慢性肝炎(CHB),而CHB是导致肝硬化和肝癌的最主要诱因。目前,针对CHB的临床治疗主要通过注射干扰素或服用核苷(酸)类似物如拉米夫定(lamivudine)、阿地福韦(adefovir)等。然而,干扰素应答率低,副作用大;使用核苷(酸)类似物易引起耐药性病毒突变株的出现。从分子水平上理解HBV的复制机制,确立新颖的药物干预靶标,并筛选与之相应的有效抑制剂,是临床实际的迫切需要,也是胡康洪博士所领导的学科组目前的主要攻关内容。
1994年,时任研究所助理研究员的胡康洪,经教育部公开选拔,获得公派赴德国留学深造的机会。在德国,他很快凭借突出的表现获得德国弗理德里希艾伯特基金会奖学金,师从著名教授Dr. D. Riesner,在杜赛尔多夫大学物理生物研究所从事类病毒的复制机理研究。在Riesner教授的影响下渐渐对病毒RNA结构和功能领域的研究产生了极大的兴趣。于是,接下来的三年,他便一心一意在弗莱堡大学从事乙型肝炎病毒分子生物学的博士后研究。在那取得一些成果后,2003年,他转战雷根斯堡大学,作为课题组长主持领导欧盟联合项目“肝纤维化的机制研究”,对乙型肝炎病毒分子生物学和肝纤维化作了深入研究,首次鉴定核因子Y-锌指蛋白267-基质金属蛋白酶10信号途径启动肝纤维化转分化过程,为设计逆转肝纤维化和治疗肝癌提供了新的药物设计靶标。从2006年4月起胡康洪在雷根斯堡大学分子和细胞解剖学研究所担任助理教授,在承担培养大学医学博士生基础课组织学和神经解剖学的一部分教学工作之余,从事肾脏发育分子生物学和干细胞组织工程研究。他所在的研究组在世界上首创用聚脂羊毛作为“人工脉管”铺设在三维培养系统中培植肾脏干细胞,使其成功分化肾小管,为肾脏发育分子生物学基础研究和利用三维培养技术开展组织工程研究作出了重要贡献。
